Correct folding proteins play critical roles in maintaining cell and body health. While some proteins will aggregation in a misfolded state resulting in diseases influenced by mutations, environments, aging or other unclear reasons. For example, several neurodegenerative diseases, cancers, and type 2 diabetes mellitus (T2DM) are caused by these misfolding or misregulated proteins. However, these potential targets are undruggable by traditional small molecular inhibitors. PROTACs emerges with great potential to eliminate these undruggable protein targets with proper binding activity, let alone for a druggable target with better safety.
