A diacylglycerol (DAG), or diglyceride, is a glyceride consisting of two fatty acid chains bonded to a glycerol molecule through ester linkages. Two main forms exist, 1,2-diacylglycerols and 1,3-diacylglycerols. DAG can be generated from phosphatidate by enzyme phosphatidic acid phosphatase in the endoplasmic reticulum. DAGs can be generated by TAG lipase in the process of lipolysis. DAG is also an important substrate that lead to the biosynthesis of triglyceride, or phosphatidylcholine and phosphatidylethanolamine. Triglyceride is important for energy storage, and phosphatidylcholine and phosphatidylethanolamine are important for cell membrane structure.
